1、試劑盒特異性因素 
1）固相載體的選擇。ELISA中常用的固相載體有微量滴定板、小珠和小試管三種，以微量滴定板zui為常見。它具有良好的吸附性能，孔底透明度高，空白值低，各板之間、同一板各孔之間性能相近。聚苯乙烯ELISA板由于原料的不同和制作工藝的差別，各產(chǎn)品的質(zhì)量差異很大。因此，在選擇試劑盒同時要對其使用的微孔板型號進行認證，評估。 
2）包被物的純度。在酶聯(lián)免疫測定尤其是間接ELISA中，試劑的特異性取決于使用抗原的純度。目前由于技術(shù)條件的限制，包被用抗原或抗體的純度不可能達到100%，所以有些非特異性顯色不可避免，只能盡可能提高純度，提高特異性。目前使用的包被抗原一般為合成多肽抗原。 
3）包被抗體的效價。具有高親和力和高特異性的包被抗體，是決定試劑特異性的重要方面。 
4 ）封閉。是ELISA中重要的一步，目的是封閉固相載體表面尚未被所占據(jù)的空隙[2]，減少后續(xù)步驟中非特異性蛋白的干擾。
