小鼠丙酮酸激酶（PK）ELISA試劑盒測定可用的標(biāo)本十分廣泛，體液(如血清)、分泌物(唾液)和排泄物(如尿液、糞便)等均可作標(biāo)本以測定其中某種抗體或抗原成份。有些標(biāo)本可直接進行測定(如血清、尿液)，有些則需經(jīng)預(yù)處理(如糞便和某些分泌物)。大部分ELISA檢測均以血清為標(biāo)本。血漿中除尚含有纖維蛋白原和抗凝劑外，其他成份均同等于血清。制備血漿標(biāo)本需借助于抗凝劑，而血清標(biāo)本只要待血清自然凝固、血收縮后即可取得。除殊情況外，在臨床研究檢驗中均以血清作為檢測標(biāo)本。在ELISA實驗中血漿和血清可同等應(yīng)用。
在它的研究、生產(chǎn)及使用中常常會碰到非特異性問題，子科生物技術(shù)部根據(jù)多年研發(fā)和臨床經(jīng)驗總結(jié)如下：
1、試劑盒特異性因素 
1）固相載體的選擇。ELISA中常用的固相載體有微量滴定板、小珠和小試管三種，以微量滴定板zui為常見[1]。它具有良好的吸附性能，孔底透明度高，空白值低，各板之間、同一板各孔之間性能相近。聚苯乙烯ELISA板由于原料的不同和制作工藝的差別，各產(chǎn)品的質(zhì)量差異很大。因此，在選擇試劑盒同時要對其使用的微孔板型號進行認證，評估。 
2）包被物的純度。在酶聯(lián)免疫測定尤其是間接ELISA中，試劑的特異性取決于使用抗原的純度。目前由于技術(shù)條件的限制，包被用抗原或抗體的純度不可能達到100%，所以有些非特異性顯色不可避免，只能盡可能提高純度，提高特異性。目前使用的包被抗原一般為合成多肽抗原。 
3）包被抗體的效價。具有高親和力和高特異性的包被抗體，是決定試劑特異性的重要方面。 
4 ）封閉。是ELISA中重要的一步，目的是封閉固相載體表面尚未被所占據(jù)的空隙[2]，減少后續(xù)步驟中非特異性蛋白的干擾。 
2、操作過程中的問題造成假陽性 
1）加樣 
對于間接ELISA標(biāo)本一般都要進行稀釋，如果加樣不準就會造成誤差，尤其當(dāng)稀釋倍數(shù)大時，很小的誤差，會導(dǎo)致較大的相對誤差，使陰性(或弱陽性)標(biāo)本呈陽性(或陰性)。加樣時應(yīng)將所加物加在ELISA板孔的底部，避免加在孔壁上部，并注意不可濺出，不可產(chǎn)生氣泡。目前，大部分血站都已使用全自動酶免加樣系統(tǒng)處理標(biāo)本，可較好地避免以上誤差。 
2）洗滌 
在ELISA中正確的洗滌是保證得到可重復(fù)結(jié)果的關(guān)健一步，應(yīng)引起操作者重視。無論是手工操作還是機器操作，得出不正確的結(jié)果常與不正確的洗滌有關(guān)，ELISA就是靠洗滌來達到分離游離和結(jié)合的酶標(biāo)記物的目的。通過洗滌以消除殘留在板孔中沒能與固體抗原或抗體結(jié)合的物質(zhì)，以及在反應(yīng)過程中非特異性吸附于固相載體的干擾物質(zhì)。 
3） 溫育 
每種試劑都有其佳反應(yīng)模式，其中溫度和溫育時間控制是重要因素。因孵育溫度高，反應(yīng)時間長，會造成整板本底高，陽性率高。溫育一般用濕盒或水浴，反應(yīng)板不宜疊放，以保證各板溫度都能迅速平衡，為避免蒸發(fā)，板上應(yīng)加蓋。 
4）酶標(biāo)儀判讀 
作為記錄測定結(jié)果的儀器，酶標(biāo)儀的性能穩(wěn)定與否，決定結(jié)果的可靠度。首先酶標(biāo)儀應(yīng)定期進行保養(yǎng)，對濾光片要定期校正；其次酶標(biāo)儀波長設(shè)置要正確，使用雙波長，一個檢測波長，一個參比波長，以消除微孔板底部劃痕、不平、指印或液面高度差異造成的光干擾。此外，在用酶標(biāo)儀讀數(shù)時先擦試微孔板底部并壓平板條。由于各種酶標(biāo)儀性能有所不同。
