小鼠腎連蛋白(NPNT)ELISA試劑盒深圳子科生物技術(shù)原理：
   (1). 抗原或抗體的固相化及抗原或抗體的酶標(biāo)記。
   (2). 結(jié)合在固相載體表面的抗原或抗體仍保持其免疫學(xué)活性。
   (3). 酶標(biāo)記的抗原或抗體既保留其免疫學(xué)活性，又保留酶的活性。
   (4). 受檢標(biāo)本與固相載體表面的抗原或抗體起反應(yīng)。再加入酶標(biāo)記的抗原或抗體，也通過反應(yīng)而結(jié)合在固相載體上。
   (5). 此時(shí)固相上的酶量與標(biāo)本中受檢物質(zhì)的量呈一定的比例。
   (6). 加入酶反應(yīng)的底物后，底物被酶催化成為有色產(chǎn)物，產(chǎn)物的量與標(biāo)本中受檢物質(zhì)的量直接相關(guān)，故可根據(jù)呈色的深淺進(jìn)行定性或定量分析。測(cè)定方法具有很高的敏感度（pg-ng/ml水平），并且重復(fù)性好。以免疫學(xué)反應(yīng)為基礎(chǔ)，將抗原、抗體的特異性反應(yīng)與酶對(duì)底物的催化作用相結(jié)合起來(lái)的一種敏感性很高的試驗(yàn)技術(shù)；

小鼠腎連蛋白(NPNT)ELISA試劑盒實(shí)驗(yàn)測(cè)定具體步驟：
1．確定本次檢測(cè)所需的已包被抗體的酶標(biāo)板孔數(shù)目，并增加 1 孔作為 TMB 空白顯色孔。 總數(shù)=樣品數(shù)+9；做雙份檢測(cè)時(shí)×2。其余重包裝好放如冰箱中。 
2．將標(biāo)準(zhǔn)品各 0.1ml 依次加入一排 7 孔中，1 孔只加樣品稀釋液的作為零孔。對(duì)于血清、體液、 組織勻漿或細(xì)胞培養(yǎng)上清，直接加已用樣品稀釋液稀釋的樣品 100μ ι 。 
3．酶標(biāo)板加上蓋，37℃反應(yīng) 90 分鐘。 
4．反應(yīng)后用自動(dòng)洗板機(jī)吸去酶標(biāo)板內(nèi)的液體；或甩去酶標(biāo)板內(nèi)液體，再對(duì)著吸水紙拍幾下。 不洗。 
5．將準(zhǔn)備好的生物素抗體工作液按每孔 0.1ml 依次加入。（TMB 空白顯色孔除 外）。37℃反應(yīng) 60 分鐘。 
6． 0.01M TBS 或 0.01M PBS 洗滌 3 次，每次浸泡 1 分鐘左右。 
7．將準(zhǔn)備好的 ABC 工作液按每孔 0.1ml 依次加入（TMB 空白顯色孔除外）。37℃反應(yīng) 30 分鐘。 
8． 0.01M TBS 或 0.01M PBS 洗滌 5 次，每次浸泡 1-2 分鐘左右。 
9．按每孔 90μ ι 依次加入已在 37℃平衡 30 分鐘的 TMB 顯色液，37℃避光反應(yīng) 20-25 分鐘 （注意：顯色時(shí)間供參考，因用戶實(shí)驗(yàn)室條件差異，顯色時(shí)間會(huì)有所不同。此時(shí)肉眼可 見標(biāo)準(zhǔn)品的前 3-4 孔有明顯的梯度藍(lán)色，后 3-4 孔差別不明顯）。 
10．按每孔 0.1ml 依次加入 TMB 終止液，此時(shí)藍(lán)色立轉(zhuǎn)黃色。 
11．用酶標(biāo)儀在 450nm 測(cè)定 OD值。

實(shí)驗(yàn)說明：
1．試劑盒保存：-20℃（較長(zhǎng)時(shí)間不用時(shí)）；2-8℃（頻繁使用時(shí)）。
2．濃洗滌液低溫保存會(huì)有鹽析出，稀釋時(shí)可在水浴中加溫助溶。
3．中、英文說明書可能會(huì)有不一致之處，請(qǐng)以英文說明書為準(zhǔn)。
4．剛開啟的酶聯(lián)板孔中可能會(huì)含有少許水樣物質(zhì)，此為正?，F(xiàn)象，不會(huì)對(duì)實(shí)驗(yàn)結(jié)果造成任何影響。
